Abstract

A new method for the determination of thyroxine in blood is described. It relies upon the quantitative dependence of the distribution of thyroxine between albumin and thyroxine-binding protein when exogenous ¹³¹I-labelled thyroxine is added to serum in vitro. Preliminary results suggest an accuracy in the estimate of the hormone of about 5–10%. Results in a group of patients whose plasma P.B.I, levels were also determined are given and shown to be similar.

The conjugate migrates by capillarity across the strip, reacting with the Ag or Ab of interest in the testing well (Fig. 3). Besides the poor sensitivity (percentage of true-positive tests in a populational sample) to low viral charge and Ab titles, IC has been the primary choice for POCT, self-test, and population triage for COVID-19 [69–71]. In the early phase of the COVID-19 pandemic, the Ab-based tests were widely used to avoid RT-PCR false-negative results in hospitalized patients late in the disease course, when the viral charge decreased.
The World Health Organizations declaration of the COVID-19 pandemic was a milestone for the scientific community. The high transmission rate and the huge number of deaths, along with the lack of knowledge about the virus and the evolution of the disease, stimulated a relentless search for diagnostic tests, treatments, and vaccines. The main challenges were the differential diagnosis of COVID-19 and the development of specific, rapid, and sensitive tests that could reach all people. RT-PCR remains the gold standard for diagnosing COVID-19. However, new methods, such as other molecular techniques and immunoassays emerged. Also, the need for accessible tests with quick results boosted the development of point of care tests (POCT) that are fast, and automated, with high precision and accuracy. This assay reduces the dependence on laboratory conditions and mass testing of the population, dispersing the pressure regarding screening and detection. This review summarizes the advances in the diagnostic field since the pandemic started, emphasizing various laboratory techniques for detecting COVID-19. We reviewed the main existing diagnostic methods, as well as POCT under development, starting with RT-PCR detection, but also exploring other nucleic acid techniques, such as digital PCR, loop-mediated isothermal amplification-based assay (RT-LAMP), clustered regularly interspaced short palindromic repeats (CRISPR), and next-generation sequencing (NGS), and immunoassay tests, and nanoparticle-based biosensors, developed as portable instruments for the rapid standard diagnosis of COVID-19.

Electrochemical biosensors have seen promising developments in the field of clinical diagnosis, food-processing quality control and environmental monitoring. In particular, nanoinspired electrochemical biosensors for monitoring biomolecular interactions occurring at the nanoscale has helped realize reagent-free, label-free, noninvasive, on site, in situ and online measurements of parameters of interest in a variety of matrices or mediums. These performance features are critical for the diagnosis of a variety of diseases of clinical interest including cancers. Cancers are known for the highest mortality rate of diseases, where current or future treatment modalities are shown to be determining factors for likelihood of a true cure. Therefore, in the quest of achieving desired diagnostic methods, electrochemical biosensors have gained wider attention. Electrochemical biosensors have been shown to provide ease of fabrication (choice of flexibility and wider tunability) and measurement using conveniently drawn samples termed as “liquid biopsies” while combating most of the challenges of contemporary methods to achieve unprecedented levels of performance (sensing ultra-trace amounts, faster detection, etc.). This chapter describes various types of electrochemical biosensors using different support materials (such as carbon allotropes, polymers, metal nanoparticles, and nanocomposites), biomolecules (such as immunoglobulins, nucleic acids, and hormones), their basic principal, applications, research trends, and future prospects of cancer diagnosis specially for point-of-care testing.

Enzyme-linked immunosorbent assays (ELISA) are effective methods for the determination of antigens in defined matrices. The method is routinely used today in clinical chemistry as well as in food analysis. It offers the possibility to measure small amounts of analytes like mycotoxins, pesticides and vitamins as well as food allergens in different food matrices. This chapter discusses key terms of the method and the main components which are required. Furthermore, it describes possible extraction procedures of the measurands from food. Finally, quality parameters for the method are discussed.
