Selective oxidation of primary alcohols mediated by nitroxyl radical in aqueous solution. Kinetics and mechanism

Abstract

The kinetics of the TEMPO-mediated oxidation of methyl α-d-glucopyranoside to sodium methyl α-d-glucopyranosiduronate were studied. An intermediate was found which was identified as the hydrated aldehyde. This was oxidised in the same manner as the alcohol, with pseudo first order rate constants ratio kobs,ald/kobs,alc — 7. The reaction mechanism is discussed with emphasis on steric factors and compared to literature data. Two different reaction pathways are postulated; under basic reaction conditions via a cyclic transition state 3 and under acid reaction conditions through an acyclic transition state 4.

    This leads to the regeneration of nitroxyl group. Oxycellulose is not reduced back to native cellulose, the same way that NaCl is not oxidized back to NaClO [146]. TEMPO-mediated oxidation can be coupled with a homogenizing mechanical treatment or sonication methods on native cellulose which has never been dried.

    Although MnO2 oxidation has been widely applied in the chemoselective oxidation of benzylic and allylic alcohols in the presence of aliphatic alcohols [19], several successful applications of TEMPO-catalyzed oxidation reaction toward this chemoselective oxidation have also been reported. Essigmann and Kobertz reported the chemoselective oxidation of the primary benzylic alcohol in 14 during the synthesis of a cis-syn furan-side photoproduct formed from a psoralen derivative and thymidine (Fig. 7) [20]. The chemoselective oxidation proceeded in the presence of primary aliphatic alcohol and phenol using oxygen, TEMPO, and CuCl to afford aldehyde 15 in 70% yield.
