Fourier Transform Infrared Spectroscopy
Sixty extracted human third molars were obtained from anonymous subjects after their signed consent under a protocol approved by the Human Assurance Committee of the Medical College of Georgia. The teeth were stored in 0.9% NaCl containing 0.02% sodium azide at 4°C for no more than 1 month. Radicular dentin devoid of enamel, cementum, and pulpal tissues was prepared from those teeth by using tungsten carbide burs, barbed broaches, and stainless steel files. FT-IR Results
Concentration-dependent and time-dependent collagen depletion by NaOCl was revealed by ATR FT-IR. Amide bands I, II, and III from the intact collagen component of mineralized dentin and phosphate and carbonate bands from the apatite component were identified in each IR spectrum. Unlike treatment with NaOCl only in which the apatite phosphate stretching vibration peak at 1020 cm−1 provides a constant intensity for normalization (12), this peak varied with sequential collagen depletion by NaOCl
Discussion
It is common for clinicians to keep the pulp chamber and root canals flooded with NaOCl during the entire period of chemomechanical preparation to maximize its instrument lubrication, tissue dissolution, and antimicrobial effects. The use of 17% EDTA as a final rinse for 2 minutes, on the contrary, is usually adequate to completely remove the smear layer from at least the coronal and middle thirds of canal walls (13).
