In 1931 von Euler and Gaddum reported the presence of a hypotensive substance in extracts of brain and intestine that could stimulate the contraction of various isolated smooth muscles. The active principle in these preparations, a peptide, was later termed substance P. In 1970  Chang and Leeman isolated a sialogogic peptide and soon thereafter characterized it as substance P by multiple biological and chemical criteria.
