Formulation and evaluation of polyethylene glycol/Xanthan gum-co-poly (Acrylic acid) interpenetrating network for controlled release of venlafaxine

Abstract

The present study aims to develop and characterize polyethylene glycol (PEG) and xanthan-gum (XG) based interpenetrating network (IPN) for the controlled release of the drug. Natural and synthetic polymers were used in combination via free radical copolymerization to synthesize series of PEG/XG-co-poly-AA IPN hydrogels. Xanthan-gum and PEG (polymers) have been cross-linked with acrylic acid as a monomer, in presence of ammonium peroxo disulfate /sodium hydrogen sulfite as initiators. For cross-linking, ethylene glycol dimethacrylate was employed. Venlafaxine HCl was incorporated in formulated hydrogel network as an experimental drug. FTIR, XRD, TGA, DCS, Sol–gel fraction, in vitro swelling, drug release, and in vivo toxicity analysis were performed to characterize the developed network. FTIR confirmed the formation of polymeric network and ensured physical entanglement of the drug within the matrix. TGA and DSC thermograms indicated that prepared hydrogels were more stable than individual constituents. Effective response for in vitro swelling and release was quantified at pH 1.2 and 7.4. No considerable toxicity was observed. It is concluded that an optimum drug delivery system can be designed to achieve desired characteristics by using a combination of polymers and altering the concentration of formulation ingredients.
